What mazdutide is — and why it matters

Mazdutide vs tirzepatide: the mechanism difference
Tirzepatide (Zepbound/Mounjaro) pairs GLP-1 with GIP — another incretin hormone that enhances insulin response and may improve tolerability. Mazdutide pairs GLP-1 with glucagon — which raises energy expenditure and specifically targets liver fat.
The practical distinction: mazdutide's glucagon component makes it particularly interesting for MASH/fatty liver disease — hepatic fat oxidation is glucagon's specialty. For pure weight loss, the two appear comparable in early data; head-to-head trials will settle it.
Side effect profile
Familiar GLP-1 territory: nausea, vomiting, diarrhea, and constipation dominate, concentrated in dose-escalation weeks. Glucagon adds a mild heart-rate increase signal (a few bpm) that investigators monitor. Trial dropout rates for side effects have been comparable to tirzepatide — the class effect, not a new problem.
When can you actually get it?
China: approved 2025 for weight management. US/EU: Lilly's global phase 3 program is running — realistic approval window is 2027-2028 if trials replicate the Chinese data. It is not available in the US now, and any "mazdutide" sold online is unregulated gray-market peptide — the same risks as unregulated retatrutide.
